Morphological and molecular variability within the fig cultivar 'Dottato' in the Italian protected designation origin area "fichi di Cosenza"

The morphological and molecular diversity among fig accessions of 'Dottato', found in the PDO "Fichi di Cosenza" area was studied by evaluating 24 morphological traits and by genotyping with 18 microsatellite markers. The microsatellite allelic profiles among the putative clones of 'Dottato' indicated a moderate genetic variability, discriminating unambiguously most of the accessions. Only two groups of identity were found. The average expected and observed heterozygosity were 0.43 and 0.62, respectively. The characterization of plant genetic resources is a precondition for genetic improvement and germplasm management. The increasing use of molecular markers for DNA-based genotype signature is crucial for variety identification and traceability in the food supply chain. We collected 75 Sicilian hazelnut accessions from private and public field collections, including widely grown varieties from the Nebrodi Mountains in north east Sicily (Italy). Seed cryopreservation of Fraxinus angustifolia Vahl

Fraxinus angustifolia is used for afforestation and for production of both valuable timber and manna , a substance with pharmacological applications. The aim of this research was to establish the optimized condition for cryopreservation of F. angustifolia seeds, in order to protect the germplasm of this species. Germination percentage and mean time of germination of non-treated seeds (control) and liquid nitrogen-treated seeds were established in water or in 10 -6 M gibberellic acid (GA 3 ). Assessment of genetic variation in SicilianHelichrysum(Asteraceae) and implication to germplasm conservation

Some taxa of Sicilian Helichrysum related to sect. Stoeachadina have an uncertain systematic position; moreover, they are rare and endangered. In order to preserve these taxa in a genebank, a strategy to keep their genetic variation as high as possible is required. Seed germination in Abies nebrodensis (Pinaceae)

Abies nebrodensis is a strict Sicilian endemic species, listed as “Critically Endangered” in the IUCN Red List of Threatened Species. Seed production highly varies annually and among specimens. The species is affected by a very low seed germination rate, mainly depending on the large number of empty seeds (from 68 to 96%). The embryo presence in seeds increases with increasing seed size and weight. Optimal germination rate (61%) was achieved at constant 5°C in the darkness. Ferula communis L. is an umbelliferous plant widespread in the Mediterranean basin and particularly abundant in Sicily and Sardinia islands (Italy). In this species, two chemotypes have been distinguished: one is poisonous and responsible of hemorrhagic syndrome of livestock, known as ferulosis, while the other one (non- poisonous) has anticancer activity due to the presence of daucane esters.
